Activity of the antiarrhythmic drug amiodarone against Leishmania (L.) infantum: an in vitro and in vivo approach J. Venom. Anim. Toxins incl. Trop. Dis.
Pinto,Erika G.; Tempone,Andre G..
Abstract Background: Considering the high toxicity and limited therapies available for treating visceral leishmaniasis (VL), the drug repositioning approach represents a faster way to deliver new therapies to the market. Methods: In this study, we described for the first time the activity of a potent antiarrhythmic, amiodarone (AMD), against L. (L.)infantum and its in vitro and in vivo activity. Results: The evaluation against promastigotes has shown that amiodarone presents leishmanicidal effect against the extracellular form, with an IC50 value of 10 μM. The activity was even greater against amastigotes in comparison with promastigotes with an IC50 value of 0.5 μM. The selectivity index in relation to the intracellular form demonstrated that the...

Antitumor activity of C-phycocyanin from Arthronema africanum (Cyanophyceae) BABT
Gardeva,Elena; Toshkova,Reneta; Yossifova,Liliya; Minkova,Kaledona; Ivanova,Natalia; Gigova,Liliana.
Pure C-phycocyanin (C-PC) was isolated from Arthronema africanumto evaluate its potential antitumor effects in vivo and in vitro. Experimental myeloid Graffi tumor in hamsters was used as a model. The cell proliferation assay showed that C-PC treatment, at concentration of 100 µg mL-1 for 24 h, significantly inhibited the growth of Graffi tumor cells (51.4% viability). Agarose gel electrophoresis of the genomic DNA of treated cells displayed time-and concentration-dependent fragmentation pattern, typical for apoptosis. Apoptotic process was related to the increase in cellular manganese and copper/zinc superoxide dismutases and glutathione reductase activities, coupled with a low catalase activity. In vivo C-PC administration (5.0 mg kg-1 body weight)...

Decreased mitochondrial metabolic requirements in fasting animals carry an oxidative cost ArchiMer
Salin, Karine; Villasevil, Eugenia M.; Anderson, Graeme J.; Auer, Sonya K.; Selman, Colin; Hartley, Richard C.; Mullen, William; Chinopoulos, Christos; Metcalfe, Neil B..
Many animals experience periods of food shortage in their natural environment. It has been hypothesised that the metabolic responses of animals to naturally‐occurring periods of food deprivation may have long‐term negative impacts on their subsequent life‐history. In particular, reductions in energy requirements in response to fasting may help preserve limited resources but potentially come at a cost of increased oxidative stress. However, little is known about this trade‐off since studies of energy metabolism are generally conducted separately from those of oxidative stress. Using a novel approach that combines measurements of mitochondrial function with in vivo levels of hydrogen peroxide (H2O2) in brown trout (Salmo trutta), we show here that fasting...

Expression and Localization of Olfactory Receptor AcerOr1 from Chinese Honey Bee, Apis cerana cerana (Hymenoptera, Apidae) BABT
Guo,Lina.
ABSTRACT Olfactory receptors are essential for recognition and detection of odor in honeybees. Although we have cloned and characterized the sequence of olfactory receptor AcerOr1 before, the tissue distribution and location of this gene in the nurse and the forager worker Apis cerana cerana were not very clear. To further investigate this information of AcerOr1 gene, we analyzed its expression and localization. The results showed that AcerOr1 mRNA was predominantly expressed in the antennae of nurse and forager bees, while the AcerOr1 protein was predominant in thorax, and its expression in antennae was higher in forager than in nurse. IHC revealed that AcerOr1 mainly localized in the olfactory neurons of the antennae. In addition, the staining intensity...

Identification of differentially expressed genes of Xanthomonas axonopodis pv. citri by representational difference analysis of cDNA Genet. Mol. Biol.
Mehta,Angela; Rosato,Yoko B..
Xanthomonas axonopodis pv. citri is a phytopathogenic bacterium responsible for citrus canker, a serious disease which causes severe losses in citriculture around the world. In this study we report the differential expression of X. axonopodis pv. citri in response to specific treatments by using Representational Difference Analysis of cDNA (cDNA RDA). cDNAs from X. axonopodis pv. citri cultured in the presence of leaf extract of the host plant (Citrus sinensis), in vivo, as well as in the complex medium were hybridized against cDNA of the bacterium grown in the minimal medium. Sequencing of the difference products obtained after the second and third hybridizations revealed a total of 37 distinct genes identified by homology searches in the genome of X....

In vivo effects of metaldehyde on Pacific oyster, Crassostrea gigas: comparing hemocyte parameters in two oyster families. ArchiMer
Moreau, Pierrick; Burgeot, Thierry; Renault, Tristan.
Pollutants via run-off into the ocean represent a potential threat to marine organisms, especially bivalves such as oysters living in coastal environments. These organisms filter large volumes of seawater and may accumulate contaminants within their tissues. Pesticide contamination in water could have a direct or indirect toxic action on tissues or cells and could induce alteration of immune system. Bivalve immunity is mainly supported by hemocytes and participates directly by phagocytosis to eliminate pathogens. Some studies have shown that pesticides can reduce immune defences and/or modify genomes in vertebrates and invertebrates. Metaldehyde is used to kill slugs, snails and other terrestrial gastropods. Although metaldehyde has been detected in...

Intraluteal Release of Prostaglandin F2α and E2 During Corpora Lutea Development in the Cow OAK
KOBAYASHI, Shu-ichi; ACOSTA, Tomas J.; HAYASHI, Kanako; BERISHA, Bajram; OZAWA, Takahiro; OHTANI, Masayuki; SCHAMS, Dieter; MIYAMOTO, Akio; 宮本, 明夫.
It is well known that prostaglandin (PG) F2α and PGE2 are actively produced by bovine early corpus luteum (CL) at higher levels than in the CL of later stages. However, there is no in vivo information about the local secretion of PGs within bovine developing CL so far. Thus, the objective of the present study was to determine in detail the real-time changes in PGF2α and PGE2 secretion within developing CL. A microdialysis system (MDS) was surgically implanted into the newly formed CL of 6 cows on Day 3 after GnRH administration (about 1.5 days after ovulation) to induce superovulation following FSH treatment. Both PGF2α and PGE2 release were high during the first 24 h within developing CL but decreased thereafter. These profiles were well reflected in the...

Seasonal changes in carbohydrate metabolism and its relationship with summer mortality of Pacific oyster Crassostrea gigas (Thunberg) in Marennes-Ole'ron bay (France) ArchiMer
Soletchnik, Patrick; Faury, Nicole; Goulletquer, Philippe.
This paper investigates the biological responses of Crassostrea gigas under traditional culture conditions on a mudflat in Marennes-Oleron bay. Summer mortality has been regularly observed in recent years in oysters reared using "on bottom" culture conditions. The present study attempts to provide a better understanding of the mortality phenomenon through biological parameters. Classical 'field parameters' such as mortality and growth rates, and quality index (dry meat weight/ dry shell weight x 1000) were monitored. Additional parameters, as biochemical composition of oyster meat and glucose incorporation capacity, were included as potential new bioindicators. The work highlighted a critical timing (May-June) preceding the summer mortality and...

Synthesis and anti-myocarditis activity in a multifunctional lanthanide microporous metal-organic framework with 1D helical chain building units BJMBR
Hong,Chenglv; Zhou,Xinlang; Huang,Weijian; Shan,Peiren; Dong,Fengquan.
A new microporous lanthanide metal-organic framework, {[Yb(BTB)(H2O) (DEF)2}n (1, DEF=N,N-Diethylformamide), with 1D nano-sized channels has been constructed by bridging helical chain secondary building units with 1,3,5-benzenetrisbenzoic acid (H3BTB) ligand. Structural characterization suggests that this complex crystallizes in the hexagonal space group P6122 and possesses 1D triangular channels with coordinated water molecules pointing to the channel center. In addition, anti-myocarditis properties of compound 1 were evaluated in vivo. The results showed that compound 1 can improve hemodynamic parameters of, and it may be a good therapeutic option for heart failure in the future.
